Oxford opened the scoring just after the hour mark as they advanced on the counterattack from a Turbines corner, with Matty Taylor slotting in at the far post following an inviting ball from the left by Ewan Clark.
The hosts sealed the win in injury time as Ewan Clarke tapped into an empty net after Jack Bearne rounded Crook on the edge of the box.
The result left Peterborough 10 points adrift with three games left of their season and join Leamington in being relegated to step three.
The final two places in the drop zone are still open - with Curzon Ashton a new contender having been sinking like a stone.
They were beaten 3-1 by Southport on Easter Monday - their 10th game without a win, having only picked up 14 points in their 19 games this year.
Hereford are currently seven points adrift but have six games to play, three more than most other clubs and still have to play Peterborough, Oxford City and Bedford.
